Trans-Trimethoxyresveratrol is a derivative of Resveratrol (RSV),and it may be a more potent anti-inflammatory, antiangiogenic and vascular-disrupting agent when compared with resveratrol. In vitro: The in vitro study of resveratrol and trans-Trimethoxyresveratrol showed rather weak cytotoxic effects on three cancer cell lines (HepG2, MCF-7, and MDA-MB-231), which contradicted a previous study reporting that resveratrol inhibited MCF-7 cells with an IC50 of about 10 μM. This discrepancy might be explained by the fact that the measurements were made 24 h after drug treatment, whereas the measurements of the previous study were taken 6 days after. The fact that the cytotoxic effect of trans-Trimethoxyresveratrol was lower than that of resveratrol is surprising, because in many studies, trans-Trimethoxyresveratrol is the most active analogue of resveratrol , although resveratrol shows much stronger antioxidant effects than that of trans-Trimethoxyresveratrol.[1] In vivo: Zebrafish embryos offer great advantage over their adults as well as other in vivo models because of the external development and optical transparency during their first few days, making them invaluable in the inspection of developmental processes. These unique advantages can even be made more useful when specific cell types are labeled with fluorescent probes. Zebrafish embryo in vivo, suggests that trans-Trimethoxyresveratrol has both more potent antiangiogenic activity and more importantly, stronger specific cytotoxic effects on endothelial cells than does resveratrol.[1]

Resveratrol-13C6 (trans-Resveratrol-13C6) is the 13C-labeled Resveratrol. Resveratrol (trans-Resveratrol; SRT501), a natural polyphenolic phytoalexin that possesses anti-oxidant, anti-inflammatory, cardioprotective, and anti-cancer properties. Resveratrol (SRT 501) has a wide spectrum of targets including mTOR, JAK, β-amyloid, Adenylyl cyclase, IKKβ, DNA polymerase. Resveratrol also is a specific SIRT1 activator[1][2][3][4]. Resveratrol is a potent pregnane X receptor (PXR) inhibitor[5]. Resveratrol is an Nrf2 activator, ameliorates aging-related progressive renal injury in mice model[6]. Resveratrol increases production of NO in endothelial cells[7].

Resveratrol (trans-Resveratrol; SRT501) is one of the numerous polyphenolic compounds found in several vegetal sources In the vast majority of cases, Resveratrol displays inhibitory/activatory effects in the micromolar range, which is potentially attainable pharmacologically, although targets with affinities in the nanomolar range have also been reported[1].
MCF-7 cells are plated in DME-F12 medium supplemented with 5% FBS in the presence of increasing concentrations of Resveratrol. Control cells are treated with the same volume of vehicle only (0.1% ethanol). Resveratrol inhibits the growth of MCF-7 cells in a dose-dependent fashion. Addition of 10 μM Resveratrol results in an 82% inhibition of MCF-7 cell growth after 6 days while at 1 μM, only a 10% inhibition is observed. The cells treated with 10 μM Resveratrol have a doubling time of 60 hr whereas control cells doubled every 30 hr. Trypan blue exclusion assay shows that at concentrations of 10 μM or lower, Resveratrol does not affect cell viability (90% viable cells) whereas at 100 μM, only 50% of the cells are viable after 6 days of Resveratrol treatment. Moreover, MCF-7 cells do not undergo apoptosis after incubation with Resveratrol at concentration of 10 μM as determined by ApoAlert Annexin V Apoptosis kit[2].
Resveratrol increases the production of nitric oxide (NO) in endothelial cells by upregulating the expression of endothelial NO synthase (eNOS), stimulating eNOS enzymatic activity, and preventing eNOS uncoupling[7].

The average tumor volume is reduced by treatment with Resveratrol (trans-Resveratrol; SRT501) at a dose of 50 mg/kg body weight (195.5±124.8 mm3; P<0.05) or 100 mg/kg body weight (81.7±70.5 mm3; P<0.001) compare with the vehicle-treated animals (315±94 mm3). There is a good correlation between the tumor volume and the tumor mass[3].

To determine the effect of Resveratrol on cell growth, MCF-7 cells are plated in 6-well plates at 105 cells per well in 2 mL of DME-F12 medium supplemented with 5% FBS in the presence or absence of increasing concentrations of Resveratrol. The cell number is measured every 2 days till day 6 with a hemocytometer after detaching the cells with trypsin-EDTA[2].

Female BALB/c (nu/nu) mice, 6 weeks old, are used. PA-1 cells (1×107 in 200 μL PBS) are injected s.c. on the right hind flank. Tumor volume (length×width×depth×0.52) is measured three times a week. After 10 days of implantation, two groups (n=10) are given Resveratrol (dissolved in 5% ethanol and 25% polyethyleneglycol 400 in distilled water) i.p. at a daily dose of 50 or 100 mg/kg body weight for consecutive 4 weeks, whereas the other group receive the vehicle only. Body weights are recorded everyday. Animals are given bromodeoxyuridine (BrdUrd; 10 mg/kg body weight, i.p.) 2 h before sacrifice. Xenograft tumors are weighed and frozen in liquid nitrogen or fixed in 10% formalin and embedded in paraffin. The BrdUrd-labeled cells in paraffin-embedded tissues are detected employing a monoclonal anti-BrdUrd antibody.

trans-Chalcone, isolated from Aronia melanocarpa skin, is a biphenolic core structure of flavonoids precursor. trans-Chalcone is a potent fatty acid synthase (FAS) and α-amylase inhibitor. trans-Chalcone causes cellcycle arrest and induces apoptosis in the breastcancer cell line MCF-7. trans-Chalcone has antifungal and anticancer activity[1][2][3].

trans-Chalcone competitively inhibits porcine pancreatic α-amylase with a Ki of 48 μM[2].
trans-Chalcone (30.23-98.03 μM; 24 hours) induces cell cycle arrest and apoptosis in MCF-7 cells[1].
trans-Chalcone (20-80 μM; 24, 48 hours) reduces the expression of the apoptosis-related protein Bcl-2[1].
trans-Chalcone (58.25 μM; 6, 24 hours) has greater inhibition of Bcl-2, induction of APAF1 and BAX, and strong induction of CIDEA in 24 hours[1].
trans-Chalcone (24 hours) inhibits MCF-7 cell viability (IC20=30.23 μM; IC50=58.25 μM; IC80=98.03 μM). trans-Chalcone (48 h) has IC50s of 41.53 μM and 48.41 μM for MCF-7 and 3T3 cell lines, respectively. trans-Chalcone exhibits a pronounced cytotoxicity activity[1].

(-)-U-50488 hydrochloride ((-)-Trans-(1S,2S)-U-50488 hydrochloride) is a selective kappa-opioid receptor (KOR) agonist (b>Kd=2.2 nM) over μ-opioid receptor (MOR) (b>Kd=430 nM). (-)-U-50488 hydrochloride is a more active enantiomer than (+) trans-(1R,2R) U-50488 (HY-15997A) or the (±) trans-racemic mixture U-50488 (HY-15997B). (-)-U-50488 hydrochloride has a potent and sustained anti-HIV effect in fected blood monocyte-derived macrophages (MDM)[1].

(-)-U-50488 hydrochloride (1 pM-100 nM; 7 days) exhibits a concentration-dependent inhibition of HIV-1 expression, the maximal inhibition at 10−13 M (approximately 73% suppression) in acutely infected blood monocyte-derived macrophages (MDM)[2].
(-)-U-50488 hydrochloride (1013 M; 7-14 days) (10−13 M) markedly inhibits HIV-1 expression both at 7 and 14 days after infection, it has a sustained inhibitory effect on HIV-1 infection in MDM[2].

(-)-U-50488 hydrochloride (intraperitoneal injection ; 5mg/kg; 2 hrs before 4% paraformaldehyde (PFA)) acutely induced pMeCP2-S421 (phosphorylation of the methyl-DNA binding protein MeCP2 at Ser421) and Fos selectively in the nucleus accumbens (NAc) but does not alter MeCP2 levels in any brain region[3].

trans-C75 ((±)-C75) inhibits PC3 cell growht with an IC50 of 35 μM at 24 h.
trans-C75 ((±)-C75)(10-50 μM) also reduces the growth of LNCaP spheroids in a concentration-dependent manner with an IC50 of 50 μM[1].
trans-C75 ((±)-C75) inhibits FAS activity and has a cytotoxic effect on tumor cell lines, without affecting food consumption.
trans-C75 ((±)-C75) inhibits CPT1 and its administration produces anorexia, suggesting that central inhibition of CPT1 is essential for the anorectic effect of C75. The differential activity of C75 enantiomers may lead to the development of potential new specific drugs for cancer and obesity[2].

(In Vivo)

C75 blocks fasting-induced c-Fos expression in the arcuate nucleus (Arc), lateral hypothalamic area (LHA), and paraventricular nucleus (PVN) 10–24 h after i.p. injection. Intraperitoneal administration of C75 at 30 mg/kg body weight inhibits food intake of mice by ≥95% within 2 h after i.p. injection[3]. C75-treated DIO mice has a 50% greater weight loss, and a 32.9% increased production of energy because of fatty acid oxidation. C75 treatment of rodent adipocytes and hepatocytes and human breast cancer cells increases fatty acid oxidation and ATP levels by increasing CPT-1 activity, even in the presence of elevated concentrations of malonyl-CoA[4].

Cell Assay

Cells are seeded in 96-well plates and incubated for 2 days to allow exponential phase growth. Cells are then ished twice with PBS and treated with C75. After 24 or 48 h incubation, MTT is added to a final concentration of 0.5 mg/ml and cultures are incubated for 2 h. Cells are then solubilized with DMSO before measuring absorbance at 570 nm. Cell growth is also measured, using MTT assay, every 24 h up to 96 h[1].

Animal Administration

Mice: C75 is administered either by i.p. (i.p.; 30 mg/kg of body weight) or i.c.v. (10 μg in 3 μL of RPMI medium 1640) injection. One, 11.5, and 24 h after i.p. injection, cumulative food intake is measured, mice are killed, brains are sectioned, and slices are subjected to immunohistochemical staining for c-Fos. All i.p. injections are given 1 h before the start of the dark cycle. For i.c.v. injection, mice are anesthetized with metofane and given 3 μl of RPMI medium 1640 (control) or C75 in RPMI medium 1640 into the lateral ventricle with a calibrated 10-μl Hamilton syringe[3].
